The treatment is a form of the poliovirus vaccine, genetically engineered to preferentially hunt down tumor cells. It is delivered straight into the brain tumor via a surgically implanted catheter. It works by killing cancer cells and simultaneously triggering an immune response against the tumor.

What phase is the Duke poliovirus study currently in?

Combining the poliovirus with other approved therapies is one approach already being tested at Duke to improve survival. A phase 2 study now underway combines the poliovirus therapy with the chemotherapy drug lomustine for patients with recurrent glioblastomas.

What is the cure for poliovirus?

There is no cure for polio, it can only be prevented. Polio vaccine, given multiple times, can protect a child for life.

Who has lived the longest with glioblastoma?

Incredibly, 2021 marks the 17th anniversary of Carmen Rice's survival from Stage 4 Glioblastoma Multiforme (GBM) brain tumor. Originally given six months to live, Carmen beat the odds to become the longest living survivor of the deadliest form of brain cancer.

What does Pvsripo stand for?

Biological: Recombinant nonpathogenic polio-rhinovirus chimera (PVSRIPO) Recombinant nonpathogenic polio-rhinovirus chimera (PVSRIPO)

Is polio curable now?

No, there is no cure for polio. Polio vaccine is the only protection against polio. Safe and effective vaccines exist – the oral polio vaccine (OPV) and the inactivated polio vaccine (IPV). OPV is administered orally and can be given by volunteers.

What countries still have polio 2020?

Polio remains endemic in two countries Afghanistan and Pakistan. Until poliovirus transmission is interrupted in these countries, all countries remain at risk of importation of polio, especially vulnerable countries with weak public health and immunization services and travel or trade links to endemic countries.

What are the final stages of glioblastoma?

Seizures occurred in nearly half of the patients in the end-of-life phase and more specifically in one-third of the patients in the week before dying. Other common symptoms reported in the end-of-life phase are progressive neurological deficits, incontinence, progressive cognitive deficits, and headache.

What triggers glioblastoma?

The causes of glioblastoma are largely unknown. However, it often occurs in people with rare genetic conditions - Turcot syndrome, neurofibromatosis type 1 and Li Fraumeni syndrome - due to mutations in a specific gene that causes many of the characteristic features of glioblastoma.

Has anyone recovered from glioblastoma?

A very small percentage of glioblastoma cases showed >3 years survival. There have been exceptional cases of long-survival spanning 10 years or more, without tumor recurrence, so as to deem those affected 'cured'.

How does pvs ripo work?

Upon intratumoral administration of recombinant oncolytic poliovirus PVS-RIPO, the poliovirus is selectively taken up by and replicates in tumor cells expressing CD155 (poliovirus receptor, PVR or NECL5) eventually causing tumor cell lysis.
