Methods: Estradiol-supplemented intact and ovariectomized immunodeficient mice were implanted with BT-474 cells. Progestin pellets were implanted simultaneously with estradiol pellets either 2 days before tumor cell injection (ie, combined) or 5 days after tumor cell injections (ie, sequentially).

Full Answer

What is the BT-474 cell line derived xenograft model used for?

The use of the human BT-474 cell line derived xenograft (CDX) model is imperative for evaluating novel cancer therapeutics, as breast cancer is the seconding leading cause of death in women. Breast cancer in humans is extremely heterogeneous, thus, any therapeutic studies performed in vitro must be followed by a xenograft study.

What is the role of xenograft in breast cancer research?

What is the BT474 cell line used for?

The BT474 cell line is used to create the CDX (Cell Line Derived Xenograft) BT474 xenograft mouse model. The BT474 breast cancer model can be used to screen anti-cancer therapeutics related to the standard of care drugs (herceptin, lapatinib, trastuzumab) or hormone-induced disease progression (progesterone, norgestrel, norethindrone and MPA).

Thank you all for your suggestions! To answer some of the questions, we inject in the mammary fat pad with 50uL cells (1x10 7) and 50uL Corning Matrigel Matrix. We have been monitoring the mice for 6 weeks now, and will continue to monitor them - hopefully they are just slow growing as others have stated.

Our lab has had succes with establishing the BT-474 xenografts ( https://www.rug.nl/research/portal/files/48622056/Complete_thesis.pdf, chapter 4). What volume of cell suspension did you use and with which matrigel? The estradiol dose seems to be the same and has not been an issue in our hands.

How long does it take to generate tumor xenografts with BT-474 and T47D cells in immunocompromised mice?

1. Introduction

BT-474 is a human invasive ductal carcinoma cell line isolated in the late 1970s from breast cancerous tissue[1]. BT-474 cells are characterised by the overexpression of epidermal growth factors receptors[2], as well as the presence of progesterone receptors and mutant p53[3].

3. Example Data

Figure 2. Brightfield micrograph showing the structure of BT-474 cells cultured for 7 days on 15 mm diameter Alvetex Scaffold discs presented in 24 well plate format. Cells were fixed, embedded in paraffin wax, sectioned (10 µm) and counterstained with haematoxylin and eosin. Scale bar: 100 µm.
